Alpha-Fetoprotein (AFP) is one kind of main glycoprotein in embryo blood serum which blong to tumor associated antigen with a molecular weight of 70kDa. AFP is synthesized in large quantities during embryonic development by fetal yolk sac and liver, but not adult. The concentration range of AFP could reach to 90~500 ng/ml in mid trimester of pregnancy. By the second year of life (about 18 months), AFP concentrations decrease rapidly, and thereafter only trace amounts are normally detected in serum.

In general, normal adults have serum AFP concentrations of less than 10ng/ml. An elevated level of AFP in serum which is higher than 10 ng/ml occurs in several malignant diseases including hepatocellular carcinoma, testicular nonseminomatous origin, and occasionally of other entodermal origin. 82% of patients with clinically verified tumors have higher amounts. AFP considerable increase in the plasma of adults is the marker of the development of hepatomacellular carcinoma or hepatitis. AFP has been used to detect early tumors in people at high risk for liver cancer. In areas where liver cancer is common, the use of AFP tests for screening has resulted in the detection of many tumors at an earlier stage. In addition, detection of elevated AFP levels can also be used in the detection of fetal open neural tube defects (NTDs).
