The FOB/TRF Rapid Test Kit is a rapid chromatographic immunoassay (noninvasive assay) for the qualitative detection of human hemoglobin and transferrin in feces specimens. lt is used as an aid in the diagnosis of gastrointestinal bleeding caused by gastrointestinal diseases such as colon polyps, colorectal carcinoma, ulcerative colitis and Crohn's disease. This assay provides only a preliminary result. Clinical expertise and professional judgment should be sought to further evaluate the result of the test.

Faecal Occult Blood (FOB) is the presence in the faeces of blood that is not clearly visible to the naked eye. This is also known as Human Occult Blood, or Human Hemoglobin. Occult blood may be the consequence of gastro-intestinal disorders such as ulcers, polyps, colitis, diverticulitis, rhagades, haemorrhoids or cancer. FOB is a important warning sign for gastrointestinal problems. The signs of these lesions are often silent in the first phases, so searching for occult blood in the stool is an important screening test, which allows to identify the presence of pathologies in the gastro-intestinal system at an early stage.

Two types of FOB tests are commercially available: guaiac dye tests and immunochemical tests (iFOBT). Traditional guaiac-based methods lack sensitivity and specificity, and also have diet restrictions prior to testing. The results of immunochemical FOB tests (iFOBT) are highly accurate and not affected by dietary peroxidases, animal blood or ascorbic acid. And dietary restriction is not necessary. A study demonstrated that immunochemical FOB screening test reduced mortality of colorectal cancer by 60%. FOB Test is a very important test method to test GI bleeding and malignant tumor of alimentary canal. It is also a regular screening test indicator of early stage tumor of alimentary canal. It is applicable to Colorectal Cancer noninvasive screening.

Transferrin is a blood plasma protein for iron ion delivery and is undetectable in normal human gastrointestinal tract. Detection of transferrin in feces or contents in the stomach indicates bleeding in gastrointestinal tract. A patient with an increased serum transferring level suffers from iron deficiency anaemia that is one of the most important symptoms when a tumour has caused chronic occult bleeding. It has been reported that fecal transferrin is elevated in patients with colorectal tumor, compared to healthy individuals. Recently, a number of proteomic studies showed that transferrin could be used as a marker expressing in a number of cancers. Unlike hemoglobin, transferrin is still detectable once released into the upper gastrointestinal tract. And transferrin is resistant to degradation by digestive enzymes and bacteria. Thus, compared to hemoglobin, transferrin is more stable in feces making transferrin an ideal marker for detecting the loss of blood from both the upper and lower intestine (gastrointestinal bleeding).

The simultaneous determination of human hemoglobin and human transferrin will be improve the accuracy and sensitivity of fecal occult blood screening test.
